What is the ibidi 8 Well Chamber, removable?

The ibidi 8 Well Chamber is a silicone chamber with eight wells bonded to a standard microscope glass slide. The defining feature is in the name: once the experiment is finished, the silicone chamber peels away and leaves the cells attached to an ordinary glass slide, ready to be coverslipped and archived like any conventional histology preparation.

That solves a specific problem. Chambered coverslips such as the µ-Slide family are excellent for culture and imaging, but the chamber is permanent — the sample stays in a plastic vessel forever, cannot be used on an upright microscope, and does not slot into a slide box. The removable chamber gives you the convenience of multi-well culture during the experiment and a standard slide afterwards.

Cultivation, fixation, staining and imaging all happen on the same slide, so cells are never transferred between vessels. That matters most with fragile or low-abundance samples, where every transfer step costs material.

The removable 8 Well Chamber appears in immunofluorescence and cell biology methods sections where cultured cells must end up on a conventional archivable glass slide, and in studies combining cell culture with upright microscopy or histology-style downstream handling.

When should I choose this over a µ-Slide 8 Well?

Choose the removable chamber when the sample needs to end up on a conventional glass slide — for archiving in a slide box, for imaging on an upright microscope, or for handing off to a histology workflow. Choose a µ-Slide 8 Well when you want the best possible optics for inverted high-resolution imaging and the sample does not need to leave its vessel.

Do I need to coat the glass before seeding?

In most cases yes. The glass surface is hydrophobic and supports little or no cell adhesion on its own. A coating such as collagen I, collagen IV, poly-L-lysine or fibronectin is standard practice, and ibidi Collagen Type I can be used for this. Robust immortalised lines sometimes attach adequately without coating, but primary cells generally will not.

When in the protocol should the chamber be removed?

After fixation and, typically, after staining — the wells are what keep reagents separated between conditions, so removing the chamber early defeats the purpose. Once the chamber comes off, apply mounting medium and a coverslip in the usual way.

Can I use it on an upright microscope?

Yes, and this is the main reason to choose it. With the chamber removed and a coverslip mounted, it is an ordinary glass slide, so it works on upright stands. Chambered coverslips such as the µ-Slide family are restricted to inverted microscopy because the chamber walls sit in the way.

Is the optical quality as good as a chambered coverslip?

For high-end techniques, no — and that is the trade-off. This product uses a standard microscopy glass slide, not a #1.5 or #1.5H coverslip, so it is not suited to TIRF, single molecule detection or super-resolution work. For routine immunofluorescence and standard fluorescence or brightfield imaging it is perfectly adequate. If you need high-NA performance, use the µ-Slide 8 Well Glass Bottom instead.
