What is the µ-Slide 8 Well high Glass Bottom?

Eight wells on a standard 25.5 x 75.5 mm slide footprint, each with 1.0 cm² of growth area and 300 µl of medium, on a No. 1.5H glass coverslip bottom of 170 µm ± 5 µm. The glass is Schott D 263 M borosilicate.

High is the operative word. The wells are 9.3 mm deep, which is what lets you run a multi step protocol, fix, permeabilise, block, several antibody incubations, several washes, without medium slopping between wells and without a single dilution error contaminating the neighbours.

Why the 1.5H glass matters

170 µm ± 5 µm is a tighter tolerance than the standard No. 1.5 rating and tighter than the 180 µm +10/-5 µm polymer bottom on 80806. High NA oil objectives are corrected for a specific coverslip thickness, and at the depth of correction TIRF and single molecule work need, a few microns of variation shows up in the image. This is the version to buy when the method is TIRF, STORM, PALM or single molecule detection.

Where I would not spend the money

For widefield fluorescence, confocal, phase contrast or DIC, the polymer bottom version, 80806, does the same job for less and its optical properties are good. Buy the glass when the objective is a high NA oil lens and the method depends on the coverslip thickness. If it does not, keep the difference. That is my view and your imaging protocol settles it.

What does 1.5H mean and why pay for it?

1.5H is glass held to 170 µm ± 5 µm, a tighter tolerance than a plain No. 1.5 coverslip and tighter than the 180 µm +10/-5 µm polymer bottom on 80806. High NA oil objectives are corrected for a specific coverslip thickness, and in TIRF or single molecule work a few microns of drift in that thickness shows up in the image.

What does high mean here?

The wells are 9.3 mm deep against a shallower well on the 8 Well. That depth is what keeps 300 µl where you put it through fixation, permeabilisation, blocking, several antibody incubations and the washes in between.

80807 or 80806?

80807 is the glass bottom, 80806 the polymer. For widefield fluorescence, confocal, phase contrast or DIC, 80806 does the same job for less. Buy 80807 when a high NA oil objective is going on and the method depends on the coverslip thickness.

Is it autoclavable?

No. Stable to 80 °C, no higher. It ships sterile, 15 or 90 individually packed or 120 at eight per tray, with a 36 month shelf life.

Is it available in Israel with local support?
