What is the Culture-Insert 3 Well?

Three silicone wells in a row, 8.4 x 12.15 x 5 mm, pre-inserted into a µ-Dish 35 mm, high with an ibiTreat surface and a #1.5 polymer coverslip bottom. Seed 70 µl per well at 3 to 7 x 105 cells/ml, wait for attachment, remove the insert. Two cell free gaps of 500 µm ± 100 µm are left, side by side in one dish.

Two gaps in one dish is the point. Seed the two outer wells differently and the middle population meets two migration fronts, both imaged in the same run under the same conditions.

Why the insert rather than a pipette tip

A scratch removes coating and leaves debris along both edges, and the gap width depends on the hand holding the tip. The moulded wells give 500 µm every time and the substrate under the gap has never been touched.

Where I would not spend the money

If one gap is enough, the 2 Well costs less and does the same thing. If you want four gaps arranged as a cross, that is the 4 Well. For many conditions rather than many gaps, the Culture-Insert 2 Well 24 in a µ-Plate 24 Well is the better buy.

How many gaps do I get?

Two, both 500 µm ± 100 µm, in one µ-Dish 35 mm, high.

What does the third well buy me over the 2 Well?

The middle well is shared. Seed the two outer wells with different cell types or different treatments and both migration fronts advance into the same central population, in one dish, in one medium. With two separate 2 Well dishes you are comparing across dishes.

Seeding volume and density?

70 µl per well at 3 to 7 x 10^5 cells/ml. Growth area is 0.22 cm² per well and coating area 0.82 cm², because the coating runs under the silicone walls.

Can I use it on my own substrate?

Not this item. This is the insert already sitting in a µ-Dish. For your own coverslip or dish, order 80369, the pack of 25 Culture-Inserts 3 Well for self insertion.

Is it available in Israel with local support?
