ibidi Culture-Insert 2 Well
Two silicone wells in a 35 mm imaging dish. Seed both, let the cells attach, lift the insert out and a cell free gap of 500 µm is left behind, with no debris along the edges and the coating under the gap untouched. A wound healing assay with the same starting geometry in every dish.

What is the Culture-Insert 2 Well?
A biocompatible silicone insert with two wells, 8.4 x 8.4 x 5 mm, already sitting inside a µ-Dish 35 mm, high with an ibiTreat surface and a #1.5 polymer coverslip bottom. Seed 70 µl of cell suspension into each well at 3 to 7 x 105 cells/ml. Once the cells have attached, lift the insert out with tweezers. What is left is a cell free gap of 500 µm ± 100 µm between two straight cell fronts.
The dish underneath is the ordinary µ-Dish 35 mm, high. Nothing gets transferred. Seed, remove the insert, add medium, put it on the inverted microscope.
Why the insert rather than a pipette tip
A scratch drags coating off the substrate and leaves a debris field along both edges, and the width of the wound depends on who held the tip. The moulded wells give the same 500 µm every time, and the surface under the gap has never been touched.
Where I would not spend the money
One gap per dish is all this gives you. If the experiment needs two gaps, buy the 3 Well. Four, buy the 4 Well. If you are counting conditions rather than gaps, the Culture-Insert 2 Well 24 goes into a µ-Plate 24 Well and gets you twelve gaps on one plate. And if the assay has to run on your own coated substrate, buy the pack of 25 inserts for self insertion instead of this one.

What gap width does it give?
500 µm ± 100 µm between the two cell fronts once the insert is out. That number holds because the wells are moulded, not because someone was careful with a pipette tip.
Can I coat the surface before seeding?
Yes. Coating area is 0.82 cm² per well against a growth area of 0.22 cm², because the coating runs under the silicone walls. Coat, wash, then seed 70 µl per well at 3 to 7 x 10^5 cells/ml.
2 Well, 3 Well or 4 Well?
The 2 Well gives one gap, the 3 Well gives two side by side, the 4 Well gives four in a cross. Same 500 µm gap and the same µ-Dish 35 mm, high in all three. If what you are counting is conditions rather than gaps, look at the Culture-Insert 2 Well 24 in a µ-Plate 24 Well instead.
Is the insert reusable?
No. It ships sterile and single use. The silicone itself is autoclavable as a material, but the µ-Dish is not, it is only stable to 80 °C.
Is it available in Israel with local support?
