iPRECIO Micro Infusion Pumps

Infusible compounds are not all the same, so the infusion method should not be either. Standard laboratory tools cover acute studies, fixed infusion rates and tethered infusion. Variable, untethered, chronic infusion has far fewer options, and that is where iPRECIO fits: an implantable pump programmed for changing rates and timing, and refilled through the skin.

iPRECIO Micro Infusion Pumps

Infusible compounds are not all the same, and the infusion method should match the compound and the study design. Acute studies, fixed rates and tethered infusion are well covered by standard laboratory tools. Variable, untethered, chronic infusion is not.

The iPRECIO Micro Infusion Pump from Primetech Corporation is implantable, programmable and refillable. Two models cover the range: SMP-310R for mice and larger animals, SMP-200 for rats and larger animals. The infusion protocol is set in software and can change rate and timing during the study, with no handling of the animal.

Infusion profiles set by the study design.

Implantable

The pump is implanted subcutaneously, so the animal moves freely with no tether during infusion, with lower stress and lower infection risk than a tethered model. Dose and timing stay under program control throughout.

Programmable

The iPRECIO software sets anything from a single constant rate to multi-step pharmacokinetic and tolerance protocols, from 0.1 µL/hr up to 30.0 µL/hr depending on the model. Typical uses:

  • Dosing time and protocol testing on small and large molecule therapeutic indices
  • Circadian infusion
  • Constant or variable dose infusion
  • Simulation of clinical dose regimens

Refillable

The reservoir is refilled through the skin via the septum port while the pump stays in place. Fluid can also be withdrawn and replaced with a different test article mid-study. With refilling, a chronic study can run up to 6 months on SMP-200 at 1 µL/hr. The elastic reservoir can be felt through the skin, a physical check on fluid level and pump function.

  • Fewer surgical interventions
  • Different concentrations or agents tested in the same animal
  • More data per animal and better welfare

What is iPRECIO and how does it differ from an osmotic pump?

iPRECIO is an implantable infusion pump for laboratory animals that is programmable and refillable. The pump sits in the subcutaneous space, so the animal moves freely with no tether during infusion. An osmotic pump delivers one fixed rate for its whole life. An iPRECIO protocol can include start and stop times, dose steps, intermittent or circadian delivery and repeat loops. The reservoir is refilled through the skin via a resealable septum, so the test article can be exchanged mid-study without further surgery.

Which iPRECIO model fits mice and which fits rats?

SMP-310R is for mice or larger animals: 24.8 x 15.0 x 7.2 mm, 3.4 g, 130 µL reservoir, 0.0 to 10.0 µL/hr in 0.1 µL/hr steps, up to 15 programmed steps. It can be reprogrammed wirelessly after implantation, at 1 to 6 m. SMP-200 is for rats or larger animals: 38.7 x 19.2 x 9.7 mm, 7.9 g, 900 µL reservoir, 0.0, 0.2, 0.5 and 1.0 to 30.0 µL/hr, up to 10 steps, programmed before implantation.

How long does the battery last?

Battery life depends on flow rate and, for SMP-310R, on how often the pump checks for wireless commands. SMP-310R with communication off runs about 67 days at 0.1 µL/hr, 41 days at 1 µL/hr and 8 days at 10 µL/hr. Checking for commands every minute cuts this to about 5 to 6.5 days. SMP-200 runs about 6 months at 1 µL/hr, 86 days at 2.5 µL/hr and 8 days at 30 µL/hr. The iPRECIO Management Software calculates battery life for a given protocol before implantation.

Which routes of administration and vehicles are supported?

Subcutaneous, intraperitoneal, intravenous (jugular or femoral), intracerebral and intracerebroventricular, and intrathecal delivery, each with the matching iPRECIO catheter. Tested vehicles include saline, PBS, DMSO up to 50% in water or saline, ethanol up to 50% in water, propylene glycol, PEG200, cyclodextrin and Solutol 15% in water, at viscosities up to 20 cP. PEG300 and PEG400 at 100% and Cremophor EL 25% in water are rated for short-term use only, 1 to 2 months.

What does a lab need to run iPRECIO, and what support is available in Israel?

Implantation is surgery, so surgical training is the main requirement for reliable results, along with a Windows 10 or 11 PC for programming. Primetech provides surgical technical notes and training videos for SC, IP, IV, ICV and intrathecal routes, a step-by-step programming guide and a vehicle compatibility test kit. NBT supports iPRECIO users in Israel with implantation guidance, protocol programming and study design.
